Stevenage and Northampton Town will be out to make up ground on the Blues.
Third-placed Stevenage, who visit Brunton Park on Saturday, can overtake the Cumbrians if they win their game in hand at home to Crewe Alexandra.
Steve Evans' side start the evening a point behind the Blues, and their inferior goal difference means only a win can put them second.
Carlisle will be hoping for another good turn from 17th-placed Crewe, who last Friday beat another promotion-chaser in Salford City.
Fourth-placed Northampton will be out to reduce their four-point gap to the top three when they host Mansfield Town.
The Cobblers will go two points behind the Blues if they beat Nigel Clough's eighth-placed Stags, who can move back into the play-off places themselves with a win.
Northampton will have played a game more than Carlisle after tonight.
Sutton United also have the chance tonight to step up their challenge.
Matt Gray's ninth-placed side, who are unbeaten in nine, are at home to Grimsby Town, and can also move into the play-off zone if they beat Paul Hurst's 16th-placed Mariners.
There are two further games in the fourth tier tonight, involving mid-table sides.
Walsall are at home to Swindon Town, while Newport County host AFC Wimbledon.
